About Aciron-800 Tablet
Aciron-800 Tablet is the high-strength oral formulation of Acyclovir 800 mg, used primarily for the treatment of herpes zoster (shingles) and severe varicella in immunocompetent adults. The 800 mg strength achieves the higher plasma levels required to effectively suppress VZV replication, which is less sensitive to acyclovir than HSV.
Like other acyclovir formulations, Aciron-800 relies on viral thymidine kinase for activation, producing acyclovir triphosphate selectively in infected cells. The active metabolite then inhibits viral DNA polymerase and is incorporated into viral DNA, terminating chain elongation and arresting viral replication.
Early initiation of high-dose oral acyclovir within 72 hours of rash onset has been shown to reduce the duration of viral shedding, accelerate lesion healing and lessen acute pain in herpes zoster. Mechanism of Action
Selective phosphorylation by viral thymidine kinase converts Acyclovir to its monophosphate; cellular kinases generate the active triphosphate. Acyclovir triphosphate inhibits viral DNA polymerase and causes obligate chain termination after incorporation into viral DNA.
Dosage & Administration
Adults: 800 mg orally five times daily (approximately every 4 hours, omitting the night dose) for 7–10 days for herpes zoster. For varicella in adolescents: 800 mg four times daily for 5 days. Adjust dose in renal impairment. Begin treatment within 72 hours of rash onset for best results.
Side Effects
Nausea, vomiting, diarrhoea, headache, fatigue, skin rash, photosensitivity. Less commonly: elevated serum creatinine, raised liver enzymes. Rare: neurotoxicity in elderly or renally impaired patients, haemolytic anaemia, thrombocytopenia.
Contraindications & Precautions
Hypersensitivity to acyclovir or valacyclovir. Use with caution in renal impairment, the elderly, dehydration, and in patients receiving nephrotoxic medications. Maintain good oral hydration.
