About Carbizoid-5 Tablet
Carbizoid-5 Tablet contains Carbimazole 5 mg, a thionamide antithyroid drug used for the treatment of hyperthyroidism, including Graves’ disease and toxic nodular goitre. The 5 mg strength is typically used for maintenance therapy once thyroid function has been normalised on higher induction doses.
Carbimazole is a prodrug rapidly converted to methimazole, which inhibits thyroid hormone synthesis by blocking thyroid peroxidase. It does not affect the release of preformed hormone, so clinical effect takes 2–4 weeks. Long-term maintenance (12–18 months) is associated with sustained remission in approximately 40% of Graves’ patients.
Carbizoid-5 is manufactured by Trumac Healthcare under WHO-GMP conditions. Available for monopoly-based PCD pharma franchise and third-party manufacturing.
Indications
- Maintenance therapy of Graves' disease and hyperthyroidism
- Toxic nodular goitre maintenance
- Preoperative thyroid preparation (with iodine)
- Pre-radioiodine therapy preparation
- Thyroid storm (adjunct, with other agents)
Mechanism of Action
Carbimazole is a prodrug rapidly converted to methimazole, which inhibits thyroid peroxidase — the enzyme responsible for iodination and coupling of tyrosine residues on thyroglobulin. This blocks the synthesis of T4 and T3 without affecting their release. Onset of clinical effect requires 2–4 weeks for preformed hormone stores to be depleted.
Dosage & Administration
Maintenance: 5–15 mg daily in divided doses, titrated to TSH and free T4. Monitor TFTs every 6–8 weeks. CBC and LFTs at baseline and as clinically indicated.
Side Effects
Rash, pruritus, mild GI upset, taste disturbance. Rare but serious: agranulocytosis (sore throat / fever — STOP and check CBC), hepatotoxicity, vasculitis, ANCA-positive vasculitis.
Contraindications & Precautions
Hypersensitivity. Severe hepatic disease. Severe bone marrow disorders. Pregnancy first trimester (use propylthiouracil instead). Bone marrow toxicity history with thionamides.
Storage
Store below 30 °C in a dry place, protected from light and moisture.
