- Recalcitrant tinea corporis or cruris

Mechanism of Action
Griseofulvin binds tubulin and disrupts the mitotic spindle of dividing fungal cells, halting fungal mitosis. It is preferentially deposited in keratin precursor cells, making newly-formed keratin resistant to fungal invasion. Mature infected keratin gradually sheds and is replaced by drug-impregnated keratin.
Dosage & Administration
- Adults: 500 mg once daily after a fatty meal
- Children: 10 mg/kg/day
- Tinea capitis: 6–8 weeks
- Onychomycosis: 6–12 months (toenails 12–18 months)
- Take with high-fat food for absorption

- Headache (common, usually transient)
- Nausea, GI upset
- Photosensitivity
- Hepatotoxicity (rare)
- Disulfiram-like reaction with alcohol
Contraindications
- Pregnancy (teratogenic) — avoid for 6 months prior to conception
- Severe hepatic impairment
