Mechanism of Action
Alfuzosin selectively antagonises post-synaptic alpha-1 adrenergic receptors in the prostate, bladder neck and prostatic urethra. This relaxes urinary tract smooth muscle tone, reduces bladder outlet obstruction and improves urinary flow without significant peripheral vasodilation, since uroselective alpha-1A/1D activity predominates over alpha-1B receptor activity.
Dosage & Administration
- Adults: one 10 mg Prolonged Release tablet once daily, immediately after the same meal each day.
- Swallow whole — do not crush or chew.
- Re-evaluate clinical response after 4–6 weeks.
Side Effects
- Dizziness, headache, fatigue, postural hypotension (especially with first dose), tachycardia, dry mouth, nausea, abdominal pain.
- Less common: syncope, intra-operative floppy iris syndrome (IFIS) during cataract surgery.
Contraindications
- Hypersensitivity to alfuzosin;
- history of postural hypotension;
- severe hepatic impairment;
- concomitant use of potent CYP3A4 inhibitors (ketoconazole, itraconazole, ritonavir).
- Inform ophthalmologist before cataract surgery.
- Not indicated in women or children.
