Mechanism of Action
Higher-dose B-complex provides therapeutic substrate for B-vitamin-dependent enzyme systems across energy metabolism (B1, B2, B3, B5), erythropoiesis and nerve function (B6, B12). Nicotinamide's role in NAD-dependent reactions supports DNA repair and is also a known sirtuin substrate. In dermatology, topical nicotinamide reduces sebum and improves barrier function.
Dosage & Administration
- Adults: 1 tablet two to three times daily for therapeutic dosing in deficiency states;
- 1 tablet daily for maintenance.
- As directed by physician.
Side Effects
- Generally well tolerated.
- Possible mild GI upset, flushing (with very high niacin doses — uncommon at therapeutic nicotinamide levels), harmless yellow urine.
- Rare hypersensitivity.
Contraindications
- Hypersensitivity to any B-vitamin.
- Caution in concurrent levodopa therapy.
- Pernicious anaemia (use injectable B12).
- Severe liver disease.
