About Trunib-5 Tablet
Trunib-5 Tablet contains Tofacitinib 5 mg, an oral selective inhibitor of Janus kinase enzymes (predominantly JAK1 and JAK3). It is a non-biologic disease-modifying anti-rheumatic drug (DMARD).
It is indicated in adults with moderate-to-severe active rheumatoid arthritis, psoriatic arthritis, ankylosing spondylitis and ulcerative colitis who have had an inadequate response or intolerance to methotrexate or other conventional DMARDs.

Mechanism of Action
Tofacitinib competitively binds the ATP-binding pocket of Janus kinases (mainly JAK1 and JAK3, with lesser activity at JAK2 and TYK2). This blocks STAT phosphorylation and downstream transcription of pro-inflammatory cytokines including IL-2, IL-6, IL-15, IL-21 and interferons. The result is suppression of T-cell activation, B-cell maturation, plasma-cell differentiation and synovial inflammation.
