About Glibicam-Forte Tablet

Glibicam-Forte Tablet combines three oral antidiabetic agents acting through complementary mechanisms: the sulphonylurea Glibenclamide 5 mg (stimulates pancreatic insulin secretion), the biguanide Metformin 500 mg (suppresses hepatic gluconeogenesis and improves insulin sensitivity) and the thiazolidinedione Pioglitazone 15 mg (enhances peripheral insulin sensitivity).

It is indicated in patients with type-2 diabetes mellitus inadequately controlled on dual oral therapy who require a third agent before initiating insulin or newer agents.

Mechanism of Action

Glibenclamide closes ATP-sensitive K+ channels on pancreatic beta cells, depolarising them and triggering insulin secretion. Metformin activates AMPK and suppresses hepatic glucose output while improving peripheral insulin sensitivity. Pioglitazone activates PPAR-gamma, increasing insulin-sensitive gene expression in adipose and skeletal muscle, reducing free-fatty-acid release.
