About Clofist-50 Tablet
Clofist-50 Tablet contains Clomifene Citrate 50 mg, a selective estrogen receptor modulator (SERM) widely used as first-line therapy for ovulation induction in women with anovulatory infertility — particularly polycystic ovary syndrome (PCOS).
Clomifene blocks oestrogen receptor feedback at the hypothalamus, leading to increased GnRH pulse frequency, FSH/LH release and follicular development. The 50 mg starting dose induces ovulation in approximately 75% of patients in the first cycle; higher doses (100–150 mg) can be used in non-responders.

Mechanism of Action
Clomifene Citrate competitively binds hypothalamic oestrogen receptors, blocking negative feedback by endogenous oestrogen. This increases GnRH pulse frequency from the hypothalamus, leading to increased FSH and LH secretion, follicular development and ovulation.
Dosage & Administration
50 mg once daily for 5 days starting on day 2–5 of menstrual cycle. If no ovulation, increase to 100 mg daily for next cycle. Maximum 150 mg/day, typically 6 cycles maximum. Specialist monitoring with ultrasound.
Side Effects
Hot flushes, ovarian enlargement, multiple pregnancy risk (~8%), visual disturbances (rare — discontinue), abdominal pain, breast tenderness, headache. Rare: ovarian hyperstimulation syndrome.

Pregnancy. Liver disease. Abnormal uterine bleeding of unknown cause. Ovarian cysts (other than PCOS). Hypersensitivity. Hormone-dependent tumours.
