Mechanism of Action
At low doses (50–300 mg/day) Amisulpride preferentially blocks presynaptic D2/D3 autoreceptors in the mesocortical and frontal pathway, raising synaptic dopamine and ameliorating negative symptoms. Selectivity for dopamine receptors and minimal binding to serotonergic, histaminic, muscarinic or adrenergic receptors limits adverse effects.
